Abstract
The invention discloses a method for acquiring explants on the basis of young shoot pinching in June and July to increase the Dangshansu pear tissue culture survival rate. The method comprises the steps that firstly, in June and July, annual thrived young shoots which grow robustly in a field and are free of pest and disease damage are selected to be pinched, the original young shoots are prompted to be induced again to generate new young shoots, and when the new young shoots grow to 10 cm-15 cm, the new young shoots are cut off to obtain the tissue culture explants; the explants are subjected to the steps of disinfecting, induction culturing, proliferation differentiation culturing, scion culturing, grafting and the like, and then Dangshansu pear regeneration plants are obtained. The method is simple, easy to operate and capable of not increasing the cost. The sufficient and good tissue culture explants also can be obtained through the measure of manual compulsive pinching in June and July to be cultivated into the Dangshansu pear regeneration plants, and the requirements of large-scale seedling propagation of Dangshansu pears or germplasm improvement research through genetic engineering are met; the survival rate of the tissue culture method reaches 96%.
