23 Jul Manufacturing method of vascularized hepatic lobule for tissue engineering
Abstract
The invention provides a manufacturing method of vascularized hepatic lobule for tissue engineering. The manufacturing method comprises the following steps: printing a compatible supportive frame of the hepatic lobule by adopting a 3D printer; inserting seven capillary glass tubes into the frame to form a forming mould of the hepatic lobule; uniformly mixing HepG2 cells and a thermo-sensitive hydrogel solution according to the cell concentration being 10<6> cells/mL, adding the obtained solution into the mould, carrying out irradiation for 30-200 s by adopting ultraviolet light, carrying out collagen cure-crosslinking, and drawing out the capillary glass tubes, thus forming a capillary channel; preparing human umbilical vein endothelial cells with the concentration being 1×10<7> cells/mL, injecting the human umbilical vein endothelial cells into the capillary channel, and carrying out culture, so that the human umbilical vein endothelial cells grow all over the inner wall of the capillary channel, and thus the vascularized hepatic lobule is obtained. The hepatic lobule prepared by adopting the method is good in biocompatibility, and can be used for the screening of liver drugs, the study on disease mechanisms, and the study on the metabolic process of the drugs in the liver.
