Inoculation method of seed cells for constructing tissue engineering nucleus pulposus of intervertebral disc

Abstract

The invention provides a seeded cell inoculation method for use in a tissue engineering of constructing intervertebral disc nucleus pulposus. A transparent hyaluronic acid is used as a carrier for the inoculation of seeded cells. The inoculation method comprises the following steps of: human or animal suspending intervertebral disc nucleus pulposus cells with the transparent hyaluronic acid; impregnating a supporting material with culture solution and absorbing the water for later use; dripping cell suspension slowly to the supporting material and keeping the supporting material stand; removing the cell-supporting material combined body into a culture flask; placing the culture solution slowly into the flask; the cell can be introduced into an animal or human body for research or treatment when the cells proliferate in the supporting material and extracellular matrixes are synthesized. The method has the advantages of improving the biological performance of the constructed tissue, the hesion rate of the seeded cells and the distribution uniformity of the seeded cells in the supporting material, along with safety, biomimetic effect, etc.
