Abstract
The invention provides an induction medium for forming malpighian cells from induced pluripotent stem cells and an induction method. The induction medium provided by the invention comprises the following components in concentration: 18-25ng/ml of an EGF, 10-20ng/ml of a b FGF, 5ng/ml of IGF-1, 0.05-0.2[mu] mol/L of dexamethasone, 0.5-2[mu] mol/L of full-transformation type vitamin A acid, 20-30ng/ml of BMP-4, 0.2-0.5mM of CaCl2, 20-35ng/ml of an SCGF, 0.6-1.0ng/ml of TGF-beta, 8-15ng/ml of a VEGF, 1.0-2.0mM of glutamine, 3-8[mu]g/ml of transferrin, (1-4)*10<-7>mol/L of triiodothyronine and (1-2)*10<-4>mol/L of adenine. By adoption of the induction medium provided by the invention, on the basis of a K-SFM culture medium, specific cell factors and complements are added, so that the probability that the induced pluripotent stem cells (ips) are differentiated to malpighian cells is increased, and seed cell sources are provided for skin tissue engineering.
