Abstract

The invention discloses an establishment method of a Dendrocalamus hamiltonii transgene system. The establishment method comprises following steps: 1) acquisition of callus tissue; 2) culturing of agrobacterium; 3) invading and co-culturing; 4) renewing culture; 5) screening and differentiation culture; 6) rooting culture; and 7) plant molecular identification. According to the invention, Dendrocalamus hamiltonii callus tissue is taken as a receptor, 10 resistant regeneration plants are successfully obtained via agrobacterium mediated transformation, transformation rate is as high as 35.7%, the 10 resistant regeneration plants are subjected to PCR molecular detection and electrophoresis product sequencing, and it is confirmed by obtained results that hygromycin phosphotransferase genes are transformed into the bamboo genome. The transformation rate of the establishment method is high; the obtained Dendrocalamus hamiltonii transgene system is stable; the establishment method is simple and convenient; effect is excellent; the establishment method possesses universality on other sympodial bamboo varieties; and solid foundation is provided for bamboo genetic engineering breeding.
