Abstract
The invention relates to the field of enzyme engineering, and discloses an enzyme-containing composition used for separating single cells and a preparation method and application thereof. The composition contains VIII type collagenase, neutral protease, a trypsin inhibitor and deoxyribonuclease, and the ratio of enzyme activity units of the VIII type collagenase, the neutral protease, the trypsininhibitor and the deoxyribonuclease is (50-90):(0.5-1):(2000-5000):1. The preparation method for the enzyme-contained composition used for separating the single cells comprises the step that the VIIItype collagenase, the neutral protease, the trypsin inhibitor and the deoxyribonuclease are mixed. The invention further discloses application of the composition in separating the single cells from apancreas sample. The method can achieve high yield and survival rate of extracting the single cells, especially the pancreatic tissue, and the problem that by adopting an existing method, the yield and survival rate of the single cells of the pancreatic tissue are lower is solved.
