Abstract
The invention discloses an efficient induction and transplanting method for tissue culture seedlings of aralia mandshrica. The efficient induction and transplanting method includes (1), acquiring aralia mandshrica leaf blades or leaf stalks with few thorns, carrying out culture on the aralia mandshrica leaf blades or leaf stalks in 2, 4-D culture media and inducing embryonic calluses; (2), maintaining and multiplying the embryonic calluses in the culture media with 2, 4-D; (3), carrying out culture on the embryonic calluses in culture media with added IBA (indolebutyric acid) to obtain maturesomatic embryos; (4), carrying out culture and germination on the mature somatic embryos in the culture media with the added IBA to obtain regeneration plants; (5), carrying out rooting and root strengthening culture on the regeneration plants; (6), transplanting the rooted tissue culture seedlings in outdoor environments. The efficient induction and transplanting method has the advantages that processes for the steps (3) and (4) are identical, accordingly, the regeneration plants can be obtained by the aid of the efficient induction and transplanting method in only two to three months, and large-scale, short-cycle, high-reproductive-rate and low-cost industrialized production can be carried out on aralia mandshrica nursery stock; the efficient induction and transplanting method further can be used as a bridge of genetic engineering, accordingly, the breeding cycle can be shortened, the forest breeding and reproductive cycles can be shortened to a great extent, and a foundation can belaid for genetic engineering improvement work for the aralia mandshrica which is a tree species.
