Abstract

The invention discloses double-rolling-circle amplification deoxyribonucleic acid hydrogel for capturing cells and a preparation method of the double-rolling-circle amplification deoxyribonucleic acidhydrogel. The preparation method comprises the following steps: preparing an RCA product 1 by using a circular DNA template 1; preparing an RCA product 2 by using a circular DNA template 2; mixing the RCA product 1 with bone marrow mesenchymal stem cells, and performing co-incubating; and adding the RCA product 2, and oscillating the material to obtain the double-rolling-circle amplification deoxyribonucleic acid hydrogel for capturing cells. The hydrogel provided by the invention has good specificity to stem cells, is an important platform for capturing cells and carrying out 3D cell culture, and can also be used for specific capture of other cells through reasonable sequence design. Moreover, the method is mild in operation process, has no obvious damage to cells, and has good application prospects in the aspects of construction of tumor co-culture models, tissue engineering, stem cell differentiation and organ regeneration.
