Abstract
The invention belongs to the biological field of tissue repair and regeneration, and in particular relates to a decellularizing method for bone materials and a method for preparing decellularized bonemeal. The decellularizing method sequentially comprises the following steps: incubation of Tris-Hcl solution, effect of trypsin solution, incubation of Tris-Hcl solution containing sodium dodecyl sulfate, washing of PBS buffer solution containing ethylenediamine tetraacetic acid and a protease inhibitor, incubation of Tris-Hcl solution containing sodium dodecyl sulfate, enzymolysis, and washing of PBS buffer solution. After the decellularizing method provided by the invention is adopted for carrying out decellularizing treatment on the bone materials, basically no residue of DNA exists, whilecollagenous fibers are reserved, for the decellularized bone materials, the extracellular matrix of natural materials is reserved, the extracellular microenvironment is good, and the biocompatibilityis good. Pig bone is extremely similar to human bone in the aspects of dissection, physiology, metabolism and bioactive molecules, and is particularly suitable for being taken as the bone tissue engineering material.
