Abstract
The invention discloses a culture method of Zhanhua winter jujube leaf calluses. The method comprises the steps that Zhanhua winter jujube leaves with 20-day leaf age are selected, the third to the fourth leaves upward from the leaf bases serve as explants, after disinfection is conducted, the explants are inoculated into induction media and placed at the temperature of 28+/-1 DEG C, dark culture or week light culture is conduced, and the winter jujube leaf calluses are obtained; the induction media contain, per liter, 1-1.5 mg of TDZ, 20 g of maltose, 10 g of agar and the balance of 3/4 MS media, and the pH is 5.8. According to the culture method of the Zhanhua winter jujube leaf calluses, the Zhanhua winter jujube leaf calluses can be obtained efficiently, the callus rate reaches up to over 85 percent, the problems of browning and vitrification do not exist, an efficient and stable leaf regeneration system is established for obtaining tissue culture seedlings and achieving rapid asexual propagation of excellent stains, and meanwhile a foundation is laid for providing excellent experimental materials for genetic engineering operation and establishing a genetic transformation system.
