Abstract
The invention belongs to the field of plant genetic engineering and particularly relates to a culture method for improving corn grain yield. The method comprises the following step: constructing a plant expression vector to transform a Zmda1-1 gene into a corn genome through a pollen tube pathway method, wherein the Zmda1-1 gene is of a mutant form of a ZmDA1 gene, which is similar to a DA1 gene in an arabidopsis thaliana plant, in the corn genome. According to the culture method disclosed by the invention, the potential of improving the corn grain yield by the Zmda1-1 gene can become a practical mature scheme by utilizing the potential property of changing the sizes of plant organs of the Zmda1-1 gene, driving the expression of the Zmda1-1 gene through a UBQI promoter and constructing the plant expression vector to transfer the Zmda1-1 gene into the corn genome; a starting vector of the plant expression vector is of a PGreen0229 plasmid vector with biosafety, is environment-friendly and can avoid a transgenic plant from affecting ecology; and according to the method disclosed by the invention, a plant expression vector solution containing target DNA (deoxyribonucleic acid) is directly transferred into corn without relying on a tissue culture regeneration system of the corn, so that an expensive instrument is not required, and the production cost is low.
