Abstract
The invention belongs to the technical field of plant genetic engineering, and particularly relates to a corn whole plant TRV carrier mediating virus-induced gene silencing method. The method comprises the following specific steps that 50 corn seeds are taken, and seed coat above embryos is ripped with a blade; seed disinfection is carried out with javelle water with available chlorine being 1% for use; amplified tomato PDS gene segments are connected into a TRV virus-induced gene silencing carrier pTRV2 through a genetic engineering method; agrobacterium-mediated transformation and culture are carried out; transformed bacterium liquid is prepared; and corn seed infecting is carried out. Corn genomes are complex, a mutant is hard to obtain, and genetic transformation is difficult and is limited by the genotype. According to the method, TRV and tomato PDS virus genes are initiatively utilized for virus carrier infecting on the corn seeds, and then corn PDS gene silencing is achieved. The method is simple, convenient to use, fast, efficient, low in cost, and needs no gene transformation, whole plant silencing is achieved, efficiency is high, and the functions of target genes in all tissue can be studied conveniently.
