Abstract
The invention discloses a construction method of a pancreatic cancer in-vitro 3D model based on a pancreatic acellular scaffold, and belongs to the technical field of tissue engineering and tumor invasion, metastasis and drug resistance. The pancreatic cancer in-vitro 3D model is constructed by simulating a tumor in-vivo microenvironment based on the pancreatic acellular scaffold and re-planting pancreatic cancer cells. On the basis of interaction of cell-cell in a traditional 2D culture medium, a cell-extracellular matrix and a three-dimensional space structure are introduced, tumor cells cangrow in the cell-extracellular matrix and the three-dimensional space structure as three-dimensional structures and are represented as circular and oval cell morphology, and changes of the nucleoplasm ratio and the nuclear fission image can be seen. The model is constructed through the following steps of (1) obtaining a pancreatic tissue; (2) treating and evaluating a decellularization effect through a hydration decellularization method; (3) carrying out cobalt-60 irradiation sterilization; and (4) re-planting the pancreatic cancer cells and evaluating the function.
