Abstract
The invention belongs to the technical field of plant genetic engineering, and in particular relates to construction and identification of rice green tissue specific expression synthetic promoters. According to the invention, some regulatory sequences, which are clear in functions and are related to the specific expression of rice tissues, are fused in different modes, and the synthetic promoters are fused with a GUS reporter gene and are introduced into a rice variety, namely Zhonghua 11. Based on transgenic rice GUS analysis, a result shows that five novel rice green tissue specific expression synthetic promoters are successfully obtained, and the synthetic promoters are different in expression intensity in different green tissues so as to meet different application purposes. The invention discloses a construction method of the synthetic promoters and a transformation vector, rice genetic transformation and a GUS tissue chemical dyeing method of transformed plants as well as a method for detecting activity of GUS protein and like processes.
