Abstract
The invention relates to the field of enzyme engineering and discloses a collagenase-containing composition and a preparation method and application thereof. The composition comprises VIII-type collagenase, neutral protease, a trypsin inhibitor and deoxyribonuclease, wherein the ratio of the enzyme activity unit of the VIII-type collagenase to the enzyme activity unit of the neutral protease to the enzyme activity unit of the trypsin inhibitor to the enzyme activity unit of the deoxyribonuclease is (100-312.5):(1-5):(5000-25000):1. The disclosed preparation method of the collagenase-containingcomposition comprises the step of mixing the VIII-type collagenase, the neutral protease, the trypsin inhibitor and the deoxyribonuclease. The invention also discloses the application of the composition in isolating single cells from a pancreatic sample. According to the collagenase-containing composition and the preparation method and application thereof, extraction of the single cells, especially pancreatic tissue, with a high yield and high activity can be achieved, and the existing problems of a low yield and low survival rate of the single cells of the pancreatic tissue are solved.
