23 Jul Cloning and identification of swine muscle specific MLC2 promoter and application thereof
Abstract
The invention belongs to the technical field of swine gene engineering and specifically relates to cloning and identification of swine muscle specific expressed gene MLC2 promoter and an application thereof. The cloning and identification process comprises the following steps of: extracting total DNA of Yorkshire pig muscle tissues, acquiring a flanking nucleotide sequence (1704bp) of swine MLC2 gene 5′ terminal by PCR amplification, constructing luciferase reporter gene plasmid of 15 MLC2 promoter’s deleted fragments with different lengths, transfecting C2C12 cell, detecting activity of the promoter of each deleted fragment, and identifying to obtain the promoter of the swine MLC2 gene, wherein the nucleotide sequence of the swine MLC2 gene’s promoter is as shown in the sequence table SEQ ID NO:1 (1704bp); and the MLC2 gene’s promoter also contains 15 promoter fragments with activities and the nucleotide sequences of the fragments are respectively as shown in the sequence table SEQ ID NO:2-16. The invention provides genetic improvement and transgenosis for pigs with important elements and means, and swine muscle tissue specificity and stability of transgenic expression can be strengthened.
