23 Jul Cloning and application of porcine skeletal muscle specific expression gene MyoG promoter
Abstract
The invention belongs to the technical field of animal gene engineering, and particularly relates to separation identification and functional verification of active regions of a porcine skeletal muscle specific expression gene MyoG promoter. The skeletal muscle specific expression gene MyoG forward 806bp promoter is cloned from the porcine genome, and the nucleotide sequence is disclosed as SEQ ID NO:1. The result proves that the promoter segment which is 806bp long has independent starting activity and muscle tissue specificity. The MyoG active promoter sequence can be used for driving the expression of the exogenous gene PPAR gamma, and can also be used for preparing a transgenic mouse. The invention also relates to a preparation method of the promoter segment, a dual-luciferase activity detection system, and application of a quantitative PCR (polymerase chain reaction) and Western Blot method in promoter activity analysis.
