Abstract
PROBLEM TO BE SOLVED: To obtain a new cDNA, capable of coding a polypeptide produced by a mantle tissue of Pinctada fucata martensii Dunker, having a specific base sequence and capable of providing a polypeptide useful for development of a new raw material for cosmetics or a new method, etc., for selecting a piece in pearl culture. SOLUTION: This new cDNA is capable of coding a polypeptide produced by a mantle epithelial tissue of Pinctada fucata martensii Dunker and used for genetic engineering production, etc., of a new polypeptide useful for developing a new raw material for cosmetics or development, etc., of a new method for selecting a piece in pearl culture. The new cDNA is obtained by isolating an mRNA from a mantle epithelial tissue of Pinctada fucata martensii Dunker, synthesizing a cDNA with a reverse transcription enzyme using the mRNA as a template, preparing a cDNA library therewith according to a conventional method, then screening the resultant cDNA library with a synthetic nucleotide capable of coding a part of an insoluble protein of a pearl layer of the Pinctada fucata martensii Dunker as a probe and recovering the cDNA from a positive clone.
