Abstract
A new-type tissue plasminogen activator (t-PA) is restructured based on gene engineering. 7 amino acid residues of HSVPVKS at positions 44-50 in F and E zonee of natural t-PA are displaced, 7 amino acid residues at positions 296-302 in t-PA structure is removed, and K1 and K2 zones in its structure are de-glycosylated to obtaine combined mutant with obviously elongated half life, increased special activity, PAI-1 resistance and affinity to fibre protein, and in turn to obtain new t-PA thrombolytic agent with improved characteristics.
