Abstract

The invention provides a method of culturing pearls by implanting nucleuses after in vitro co-culture of mantle free cells and the tissue engineering scaffold pearl nucleuses. The method comprises the steps of separating mantle tissue of pearl shells into single existent free cells through the enzyme digestion method and preparing a cell suspension with a certain cell density; sticking a layer of a high-molecular polymer tissue engineering scaffold with basically uniform thickness to the surface of a pearl nucleus which is to be implanted in a pearl mussel and culturing the pearl nucleus and the free pearl shell mantle cells in vitro together to make the mantle cells cling to the fibers of the tissue engineering scaffold at the surface of the pearl nucleus for mitosis and growth; after the cells grow and propagate in the tissue engineering scaffold and form a pearl sac with cells basically uniformly distributed around the surface of the pearl nucleus, transplanting the pearl nucleus into the body of the pearl mussel; culturing pearls through culture in a pearl culture farm. The method solves the problem of traditional mantle cell piece implantation method that cells are not distributed uniformly after migration around pearl nucleuses and thus cause strangely-shaped pearls, and guarantees great compatibility of pearl nucleuses and pearl mussels.
