Combined mutant of histotypic plasminogen activator

Abstract

Combined mutant, named rPA, of new tissue-type plasminogen activtor, named t-PA, is constituted through protein engineering of altering the molecular structure of t-PA, deleting the EGF area, Finger area and Kringle I area while retaining the Kringle II area and proteinase area with thrombolytic effect, and mutating the amino acid HRR in 126-128 positions of t-PA molecule into AAA. The cDNA of rPA is inserted into the yeast expressing plamid to convert Pichia yeast, the high-expression engineering saccharomycete is obtained through screening, and through further expression and serial purification steps, high-purity and high-specific activity rPA is prepared. The rPA has the advantages of natural t-PA, such as high-specific activity rPA is prepared.
