23 Jul Tissue engineering reconstruction method for human corneal endothelium
Abstract
The invention relates to a method for human corneal endothelium reconstruction of tissue engineering, which comprises the following steps: the corneal endothelial cells is cultured in vitro to the logarithmic phase by using 20% of calf serum-DMEM/F12 culture solution, amniotic carrier scaffolds with epithelial layer completely removed is obtained by the tryptic digestion, the scaffolds is tiled in the holes of a culture plate and after being firm and dry-bonded, the corneal endothelial cells in the logarithmic phase and containing IV type collagen and DMEM/F12 culture solution of 20% of calf serum are inoculated in the holes of the culture plate tiled with the amniotic carrier scaffolds to start culture in vitro and the human corneal endothelium reconstruction in vitro of tissue engineering is carried out. The invention has scientific and rational process; the reconstructed human corneal endothelium of tissue engineering is capable of being used for mass production, thus meeting the great demand of the corneal endothelium blind clinical transportation treatment on human corneal endothelium of tissue engineering; the human corneal endothelium reconstruction in vitro of tissue engineering and clinical treatment have low cost.
