Tissue engineered cardiac muscle using OPF (Oligo(Poly(Ethylene Glycol) Fumarate) hydrogel as a carrier and preparation method thereof

Abstract

The invention belongs to the field of tissue engineering and regenerative medicine, and discloses an injectable tissue engineered cardiac muscle which uses low oligo(poly(ethylene glycol) fumarate (OPF) hydrogel as a support material and contains OPF hydrogel and iPS (Induced Pluripotent Stem) cells or cardiac muscle cells originating from the iPS cells. A preparation method of the tissue engineered cardiac muscle specifically comprises the steps of: adding the OPF and a crosslinking agent into a PBS (Poly-Butylene Succinate) solution; then adding an ammonium persulfate solution and a TEMED (Tetramethylethylenediamine) solution; and adding PBS suspension containing the iPS cells and slightly and uniformly mixing. The invention also discloses the preparation method and application of the tissue engineered cardiac muscle. In the invention, by adopting the OPF hydrogel as a carrier, the detension ratio and the survival ratio of the iPS cell are improved and a good repairing effect on heart functions of an infarcted zone is good; secondly, the tissue engineered cardiac muscle disclosed by the invention can be injected so that the operation is convenient; and in addition, because the iPS cells are used as seed cells, the problems on ethical morals and immunological rejection do not exist. In the invention, the preparation method is simple and the implementation conditions are mild.
