Abstract

The invention belongs to the technical field of genetic engineering, particularly relates to a shRNA (short hairpin ribonucleic acid) transgene recombinant plasmid capable of inhibiting a foot and mouth disease virus and an application thereof. The plasmid comprises two shRNA expression genes which can be used for simultaneously coding two kinds of specific shRNAs which respectively target 3D RNA polymerase of a foot and mouth virus genome and 2B non-structural protein conserved domains, wherein the two shRNA expression gens are in a serial connection state. The plasmid is applicable to various transgenosis technologies. After the plasmid is transformed into a porcine cell IBRS-2 which is cultured in-vitro by using a PB transposon system, challenge assays prove that: a screened transgene cell has the capability of resisting 20-50 TCID (tissue culture infectious dose) 50 dose of O-type or Asian I-type FMDV (foot and mouth disease virus) strain infection. After the plasmid is introduced into fertilized eggs of mice through microscopic injection, the survival rate of screened transgene mouse families is remarkably improved relative to mice in a same brood when the mice is infected by a 3-6LD50 O type FMDV strain.
