Abstract

The present invention discloses a goat skeletal muscle-specific gene actin promoter-driven Fat-1 expression vector and a construction method thereof, and the expression vector is constructed as follows: first, Fat-1 gene is cloned into pEGFP-N1 vector to construct pEGFPN1-Fat-1 vector, CMV enhancer obtained by PCR amplifying is inserted into the promoter and enhancer-free pEGFPN1-Fat-1 vector to construct pEGFPN1-Fat-1-Ehancer vector, and finally skeletal muscle-specific gene alpha-actin promoter is inserted into the pEGFPN1-Fat-1-Ehancer vector to construct alpha-actin promoter-pEGFPN1-Fat-1 expression vector. The expression vector has high activity while maintaining muscle tissue specificity, and lays a foundation for future applications in genetic engineering.
