Preparation method of multi-scale three-dimensional biological tissue engineering scaffold

Abstract

The invention discloses a preparation method of a multi-scale three-dimensional biological tissue engineering scaffold. The preparation method comprises the following steps: (1) making a mode having a three-dimensional grid structure or cavity structure; (2) making a two-dimensional fiber thin film through electrostatic spinning; (3) simultaneously adding the mold and the two-dimensional fiber thin film to a liquid-phase system which is arranged on the inner surface of the mold, has an affinity abnd is capable of generating bubbles, and meanwhile, keeping top surface of the two-dimensional fiber thin film pressed by the mold, so that under a combined action of the bubbles and capillary force, the two-dimensional fiber thin film is promoted to migrate and expand into grid holes or cavities of the three-dimensional grid structure, and the two-dimensional fiber thin film is shaped in the three-dimensional grid structure; and (4) conducting vacuum drying, so that the tissue engineering scaffold which has a guidance effect on cells is obtained; and optionally, dismantling the mold. According to the preparation method disclosed by the invention, the three-dimensional nano-fiber scaffold, which is represented in a controllable geometrical form, is prepared by integrating the rapid and high-precision personalized mold-making of 3D printing as well as advantages of the electrostatic spinning in preparing high-porosity nano-fibers; and the preparation method is simple in process and strong in practicability.
