Abstract
The invention relates to a method for detecting and locating leifsonia xyli subsp.xyli LXX. The method comprises frozen sectioning, pretreatment, in-situ PCR, immunohistochemistry, and observation and detection. Compared with a mode of researching LXX planting through sugarcane converting by constructing engineering LXX with a GFP (Green Fluorescent Protein), the method for detecting and locating the LXX can carry out indirect in-situ PCR for locating a specific nucleic acid of the LXX in a frozen section of a sugarcane infected plant infected with ratoon stunting disease, realize detection of the LXX quickly, and research tissue localization of the LXX; furthermore, the problems of low efficiency, high consumption, high technical requirements and the like which are caused by construction and conversion of engineering bacteria are not considered. The method disclosed by the invention effectively avoids the problems of relatively high difficulty in in-vitro pure cultivation of the LXX, relatively low multi-resistance specificity, relatively high difficulty in preparation of a monoclonal antibody and the like at the present, and successfully realizes quick and accurate detection and localization of the LXX.
