23 Jul Experimental research method for repairing full-thickness skin defects of nude mice through tissue engineering skin
Abstract
The invention discloses an experimental research method for repairing full-thickness skin defects of nude mice through the tissue engineering skin. The method comprises the following steps that firstly, healthy male nude mice are divided into two groups, namely the tissue engineering group and the blank control group, and each group includes 10 nude mice; secondly, after the nude mice are narcotized, the center of the back of each nude mouse is cut into a circular defect with the diameter about 1 cm; thirdly, the built hAECs-DED tissue engineering skin is used for covering wound surfaces of the mice in the tissue engineering group at the defects of the nude mice, and nothing covers wound surfaces of the mice in the blank control group; fourthly, observation of postoperative wound surface healing conditions and histological detection are conducted; fifthly, sampling is conducted on the back operation areas of the nude mice four weeks after operations, and the tissue is sectioned and stained with HE; sixthly, statistical analysis is conducted. According to the method, the built hAECs-DED tissue engineering skin is transplanted to the wound surfaces of the nude mice, the effect that the tissue engineering skin product is used for repairing the full-thickness skin defects of the nude mice is detected and learnt in the aspects of wound surface healing rate, wound surface healing time, histopathology and the like, and an experimental basis and a technical support are provided for future clinical application.
