Abstract
The invention relates to an isolated culture method and application of cells and in particular relates to an isolated culture method and application of adipose mesenchymal stem cells. The isolated culture method comprises the following steps: taking a human adipose tissue and digesting by utilizing collagenase; carrying out gradient centrifuging and filtering; then carrying out amplification culture in an in-vitro culture system, wherein the in-vitro culture system contains DMEM (Dulbecco’s Modified Eagle Medium)/F12, EFG (Epidermal Growth Factor) and FCS (Fetal Bovine Serum). The adipose mesenchymal stem cells provided by the invention have phenotypes of CD11a<->, CD31<->, CD34<->, CD45<-> and HLA-DR<->, and CD44<+> or F1k-1<+>. The invention further discloses novel medical application of the adipose mesenchymal stem cells and a new selection is provided for seed cell sources in tissue engineering and regeneration medicine and application of adult stem cell groups.
