23 Jul Method for establishment of efficient and stable regeneration system with dendrocalamus asper shoot tips
Abstract
Belonging to the technical field of plant tissue culture, the invention relates to a method for establishment of an efficient and stable regeneration system with dendrocalamus asper shoot tips. The method is carried out by the steps of: 1) acquisition of aseptic buds; 2) callus induction culture; 3) callus differentiation culture; 4) rooting culture of regenerated seedlings; and 5) transplantationof regenerated plants. The method provided by the invention can achieve a highest callus induction rate of 91.7%, wherein the milky white compact granular callus induction rate is 61.7%, a highest seedling differentiation rate is 46.67%, and a highest rooting rate is 55%. The method can enable an explant to induce callus efficiently and establish an efficient and stable regeneration system successfully, creates favorable conditions for bamboo genetic transformation and cell engineering research, and lays a good foundation for breeding with transgenic technology. The method has the characteristics of simple and practicable implementation steps, loose implementation conditions, and remarkable effect.
