Abstract
The invention discloses double rolling circle amplification (RCA) DNA hydrogel and a preparation method thereof. The preparation method comprises the steps as follows: adding sterile water to a circular DNA template I, phi 29DNA polymerase, a phi 29DNA polymerase buffer solution, BSA, dNTPs and NaCl; performing oscillation to obtain an RCA product 1; adding sterile water to a circular DNA templateII, phi 29DNA polymerase, a phi 29DNA polymerase buffer solution, BSA, dNTPs and NaCl; performing oscillation to obtain an RCA product 2 complementary with the RCA product 1 sequence; and mixing theRCA products 1 and 2, and performing deactivation, mixing and oscillation to obtain the double RCA DNA hydrogel. The method is short in synthesis time. The hydorgel has good biocompatibility and highdesignability, and the material is endowed with more functional characteristics by design of temperate. The hydrogel can be applied to drug delivery, cell culture and tissue engineering.
