Abstract

The present invention relates to a method for preparing hydrogel based on decellularized tissue using a supercritical fluid-organic solvent system, and to a technique for application of the hydrogel as a material for tissue engineering having improved angiogenesis and tissue regeneration abilities. More specifically, the method comprises the steps of: (a) performing decellularization by contacting biological tissue with a supercritical fluid and an organic solvent at the same time; (b) washing the decellularized tissue; (c) manufacturing a decellularized tissue solution by mixing the washed decellularized tissue with pepsin and an acidic solution; (d) treating the decellularized tissue solution with a basic solution to titrate the same at pH 5.5 to 7.8; and (e) leaving the titrated decellularized tissue solution at 30 to 40°C. According to the present invention, decellularized hydrogel with maximized preservability of various tissue-derived proteins, growth factors, and cytokines, is prepared.
